Clinical Evaluation:

A healthcare provider will assess the patient's symptoms, medical history, and physical examination findings. Specific signs and symptoms associated with Staphylococcus aureus infections, such as skin lesions, fever, or respiratory symptoms, may raise suspicion for the presence of the bacterium.


Laboratory Tests:

 
Culture and Sensitivity Testing: 
A sample of tissue, fluid, or discharge from the affected area (e.g., wound swab, blood sample) is collected and sent to a laboratory for culture. In the case of suspected skin or soft tissue infections, a swab may be used to collect a sample from the affected site. 
 
Blood cultures are typically obtained for suspected bloodstream infections. The specimen is cultured on appropriate media to allow the growth of bacteria, including Staphylococcus aureus. Once grown, the bacteria can be identified, and susceptibility testing can determine which antibiotics are effective against the specific strain.
 
Gram Staining: 
Gram staining of clinical specimens can provide preliminary information about the morphology and arrangement of bacteria. Staphylococcus aureus appears as Gram-positive cocci in clusters under the microscope.
 
PCR (Polymerase Chain Reaction): 
Molecular diagnostic tests, such as PCR, can rapidly detect the presence of Staphylococcus aureus DNA in clinical samples. PCR assays are often used for rapid identification of MRSA strains or in cases where traditional culture methods may be delayed or inconclusive.


Imaging Studies: 
In cases of suspected deep-seated infections, such as osteomyelitis or septic arthritis, imaging studies such as X-rays, ultrasound, CT scans, or MRI scans may be performed to assess the extent of tissue involvement and aid in diagnosis.


Other Tests: 
Additional tests may be performed depending on the clinical presentation and suspected source of infection. These may include serological tests for specific antibodies, toxin assays for certain syndromes like toxic shock syndrome, or molecular typing methods for epidemiological purposes.
